Piero Capelli ha studiato filologia classica, orientalistica antica ed ebraistica all'Università di Pisa (laurea in Lettere Classiche, 1987), alla Scuola Normale Superiore di Pisa (diploma di licenza in Lettere, 1987; frequenza al corso di perfezionamento in Lettere, 1987-1990; borsista della Fondazione Giorgio Pasquali, 1994-1995) e nelle università di Torino (dottorato di ricerca in Ebraistica, 1995; borsista post-dottorato,1996-1997) e Vienna (borsista del Bundesministerium für Wissenschaft und Forschung della Repubblica Federale d'Austria, 1993 e 1995). È stato docente a contratto di ebraico biblico e rabbinico all'Istituto di Scienze Religiose di Trento (1998-1999) e all'Istituto Universitario Orientale di Napoli (1999). A partire dal 2000 è stato prima docente a contratto, poi ricercatore, poi professore associato, e infine professore ordinario di ebraico a Ca' Foscari. Nel Trinity Term 2005 e negli Hilary e Trinity Terms 2011 è stato Visiting Scholar presso l'Oxford Centre for Hebrew and Jewish Studies. Dall'ottobre 2007 al maggio 2009 è stato Visiting Scholar-in-Residence presso il Rabbi Donald A. Tam Institute for Jewish Studies della Emory University di Atlanta. Dal 2011 al 2016 è stato Visiting Associate Professory in History presso la Zanvyl Krieger School of Arts and Sciences della Johns Hopkins University di Baltimora. Nel 2012-2013 è stato Primo Levi Fellow presso lo Herbert D. Katz Center for Advanced Judaic Studies della University of Pennsylvania a Philadelphia. Nel 2014 è stato vincitore di una Franklin Research Grant della American Philosophical Society di Philadelphia. Nel 2016 è stato vincitore di una Bi-National Scholarship for Scientific Cooperation between Italy and Israel, grazie alla quale nel semestre primaverile 2017 ha condotto ricerche presso il Center for the Study of Conversion and Inter-Religious Encounters della Ben-Gurion University of the Negev di Beersheva.
Il suo campo d'indagine è la storia dei testi e delle idee nell'ebraismo tardoantico e medievale, in particolare nei testi rabbinici in ebraico e in aramaico e negli apocrifi e pseudepigrafi in greco. Si sta occupando della prima edizione critica del resoconto ebraico del primo processo pubblico contro il Talmud, tenuto a Parigi nel 1240.
È direttore di «Henoch» (rivista internazionale di studi storico-testuali su giudaismo e cristianesimo in età antica e medievale). È membro dei comitati scientifici della rivista di cultura «Humanitas» e della rivista internazionale dell'Associazione Italiana per lo Studio del Giudaismo (AISG) «Materia Giudaica», e del comitato di redazione della rivista «Annali di Ca' Foscari. Serie orientale». Dal 2020 è socio corrispondente della Classe di Lettere e Arti dell'Accademia Roveretana degli Agiati.
